RQ-HPV HR-LR Multiplex - A Testing Solution for 22 HPV Types
Human Papillomavirus (HPV) is the leading cause of cervical cancer, and WHO recommends HPV DNA testing as the primary screening test. But how should you choose an HPV DNA test kit? This article will give you more information to help you choose.

An Overview of HPV
Human Papillomavirus (HPV) is an extremely common sexually transmitted DNA virus and is known to be the leading cause of cervical cancer. Just over 40 HPV types can infect the genital area. Based on their association with cervical cancer, HPV types are divided into 2 groups: high-risk HPV and low-risk HPV.
More than 99% of cervical cancer cases are positive for high-risk HPV, and about 70% of cancer cases are associated with 2 types, HPV-16 and HPV-18. Although low-risk HPV usually causes only benign growths, about 90% of anogenital warts are caused by 2 types, HPV-6 and HPV-11.

Choosing an HPV DNA Test Kit
According to WHO recommendations, HPV DNA testing using nucleic acid amplification tests (NAAT) should be chosen as the primary cervical cancer screening test. Real-time PCR is currently the most widely used NAAT technique.
However, a major challenge today is that there are many different HPV DNA test kits using real-time PCR. So how can you choose a kit that is both high-quality and suited to your needs?
To choose a real-time PCR kit for HPV DNA testing, we need to consider the following aspects:
Sensitivity and Specificity:
These parameters represent the accuracy of a product and will differ between kits and between manufacturers.
HPV Genotyping Capability:
More than 40 HPV types can infect the genital area. Detecting and differentiating HPV types (or groups of HPV types) can help explain the patient's condition.
Sample DNA Input:
The DNA volume of a sample is limited, while in practice it is sometimes necessary to run many different tests on the same sample. Saving DNA can make a sample sufficient for more tests.
Preventing Contamination Incidents:
Contamination of a PCR laboratory is extremely serious: PCR products that accidentally escape from a tube will contaminate the entire area, causing widespread false positives. Remedying the consequences of such an incident is also extremely difficult.
To prevent this, some PCR kits today use a dUTP/UNG system to limit the impact of residual DNA and to make it easier to resolve incidents.
Compatibility with Multiple PCR Instruments
A laboratory sometimes has several PCR instruments from different manufacturers. An assay that is compatible with multiple instrument models is a plus.
Result Interpretation Software
Reviewing the signal of each sample to analyze results is very time-consuming and subjective. Software that helps conclude whether a sample is negative/positive saves a great deal of time, especially for laboratories with large sample volumes.
Viet Huy's HPV DNA Testing Solution

Figure 1: RQ-HPV HR-LR Multiplex product (AB analitica, Italy).
REALQUALITY RQ-HPV HR-LR Multiplex (AB analitica, Italy) is a CE-IVD real-time PCR kit line that detects 22 HPV types simultaneously, with genotyping of HPV-16, 18, 6, and 11. The additional genotyping of HPV-6 and HPV-11 allows clinicians to explain the cause of 90% of genital wart cases.
The product aims to optimize cost and handling while maximizing the ability to test for HPV and other sexually transmitted infection (STI) pathogens simultaneously, thanks to 3 key factors:
- - Reduced DNA input.
- - Fewer master mixes.
- - A thermal profile identical to that of the REALQUALITY STI kit line, allowing real-time PCR for HPV to be run simultaneously with up to 7 other sexually transmitted infection (STI) pathogens.
In addition, REALQUALITY RQ-HPV HR-LR Multiplex also aims to optimize the user experience with 3 conveniences:
- - A dUTP/UNG system in the master mix decontaminates residual PCR products from previous PCR runs and makes it easier to resolve contamination incidents in the PCR laboratory.
- - An automated version is available on the GENEQUALITY 120 system (AB analitica, Italy).
- - AB Genius Report result analysis software.
In summary, compared with other HPV testing solutions, REALQUALITY RQ-HPV HR-LR Multiplex offers the following convenient features:
| RQ-HPV HR-LR Multiplex | Most other manufacturers |
| Detects 22 HPV types | Detects 14 HPV types |
