HLA-B27 TEST KIT USING THE REAL-TIME PCR TECHNIQUE

HLA-B27 is a factor associated with many autoimmune diseases and ankylosing spondylitis. Real-time PCR testing is a method that determines HLA-B27 status quickly and accurately, with high specificity and sensitivity.

Performance Characteristics of the HLA-B27 Real-TM Kit

HLA-B27 Real-TM is an IVD test kit for detecting HLA-B27 by the Real-time PCR method, compatible with common Real-time PCR instruments on the market.

Sample Source:

Whole blood in EDTA tubes; samples can be stored at 2-8℃ for up to 48 hours, or at -20℃ to -80℃ for longer storage.

Extraction:

Any commercial extraction kit can be used to obtain human genomic DNA from whole blood samples. The DNA concentration for optimal test performance is 20-200 ng/μL.

The reaction includes negative and positive controls that monitor the PCR process in order to reach the most accurate conclusion about the status of the patient sample.

• The negative control checks for contamination with target DNA in reagents, equipment, and samples, thereby correctly determining whether a sample is truly positive. 
• The positive control checks the specificity of the primers for the target agent and verifies the performance of the Real-time PCR reagents.  
• In addition, the PCR reaction also amplifies the beta-globin gene present in human cells, which serves as an endogenous internal control to monitor PCR inhibitors and prevent false-negative results.

The HLA-B27 status of the sample is assessed based on the difference between the fluorescence signals (Ct) of the FAM channel and the HEX channel. HLA-B27 is detected in the FAM channel, and the IC (beta-globin gene) is detected in the HEX channel.
